• Supplemental Figure - In this experiment, we tested the specificity of the antiserum on native proteins by immunostaining of MDA MB 436 cells overexpressing NF1 C2 and NF1 C5. MDA-MB 436 cells were transiently transfected with expression plasmids for NF1-C2 and NF1-C5 proteins. The cells were then immunostained with either the NF1-C specific (8199) antibody or the pNF1-C2 specific antiserum. Immunostaining with the NF1 C specific antibody (8199) detected nuclear proteins in cells overexpressing both NF1 C2 and NF1 C5, while the new pNF1 C2 antiserum only detected proteins in cells overexpressing NF1 C2. This demonstrates that the pNF1-C2 specific antiserum was specific for native NF1-C2 proteins. This is in agreement with the results obtained by denatured proteins on Western blot. Note that this supplemented figure needs to be printed on a good quality printer in order to be informative in print.
