• Supplemental Figure 1 - TRα1 and TRβ1 expression in individual HepG2 transformant clones, and validation of microarray data by reverse transcriptase PCR analysis. A, mRNA levels for TRα1 were examined from two independent transformant clones of HepG2- TRα1 cells, and for TRβ1 from two independent transformant clones of HepG2-TRβ1 cells; mRNAs from the corresponding pools of HepG2-TRα1 or HepG2-TRβ1 transformants employed in the microarray analysis were isolated and analyzed in parallel. All cells were treated minus or plus T3 for 6 hrs and analyzed by rtPCR as in manuscript Figure 6. B, The mRNAs from the same independent clones in Fig. S1A were subjected to rtPCR analysis to determine the expression of the target genes indicated, using appropriate primers. The resulting PCR products were stained with Sybr Green and the gel band intensity values were quantified using an alpha-Innotech FluorChem 8900 gel documentation system as in manuscript Figure 6. The level of expression of each gene product was defined as “1” for the empty-vector control in the absence of T3 (mean + S.E.M., n = 3). C-D, The mRNAs from TRα1, TRβ1, or empty plasmid (NR) transformants, minus or plus 6 h. of T3 treatment, were analyzed for the target genes indicated. Methodology was as in manuscript Figure 6.
