• Supplemental Figure 1 - Certain, but not all, RCCC TR mutants are impaired in T3 binding. The radiolabeled TRs indicated were incubated with increasing concentrations of T3 and 0.05U elastase in a protease protection assay. The samples were analyzed by SDS-PAGE and the protease-resistant cores (indicated by bars to left of each panel) were visualized by phosphorimager analysis. Three or more independent experiments were performed; an image of a representative experiment is shown.
• Supplemental Figure 2 - The different CoRNR boxes in SMRT and in NCoR display similar release from TRs in response to T3. (A-D) The ability of wt-TRα1 and M388I-TRα1 to bind to GST fusions representing the individual SMRT and NCoR CoRNR boxes was determined in the presence of increasing amounts of hormone, as described in Figure 5. Values shown are relative to the binding seen for each TR in the absence of T3 (defined as 100) to facilitate comparison of the EC50 values.
